Comfy mice lead to better science: Are cold mice affecting drug testing?

ScienceDaily (Mar. 30, 2012) ? Nine out of 10 drugs successfully tested in mice and other animal models ultimately fail to work in people, and one reason may be traced back to a common fact of life for laboratory mice: they're cold, according to a researcher at the Stanford University School of Medicine.

Laboratory mice, who account for the vast majority of animal research subjects, are routinely housed in chilly conditions, which may affect their well-being as well as the outcome of research studies, said Joseph Garner, PhD, associate professor of comparative medicine.

"If you want to design a drug that will help a patient in the hospital, you cannot reasonably do that in animals that are cold-stressed and are compensating with an elevated metabolic rate," Garner said. "This will change all aspects of their physiology -- such as how fast the liver breaks down a drug -- which can't help but increase the chance that a drug will behave differently in mice and in humans."

In a new study, Garner and his colleagues report finding an easy solution to the problem: Simply provide the animals with the proper materials, and they'll build a cozy nest that allows them to naturally regulate their temperatures to a comfortable level. These thermally content mice would be more physiologically comparable to humans and thus might serve as more meaningful research subjects, Garner said.

"Why not let them do what they do in the wild, which is build nests? Mice can happily infest a meat freezer, with temperatures far below zero, but they survive and breed because they build these wonderful nests," he said.

The study, part of nearly seven years of work with mouse nesting behavior, is the first to "ask" mice to rate the value of nesting material in terms of temperature savings, which is an important first step in setting standards for nesting material, said Garner, whose work has focused on the well-being of the mouse. He is the senior author of the study, which was published online March 30 in PLoS ONE.

Mice, which Garner calls "one of the most fantastic animals on Earth," have evolved in the same environment as humans for thousands of years, making them remarkably adaptable, able to live virtually anywhere. For that reason, they make excellent research subjects, with hundreds of millions of them populating laboratories around the world.

Given the option, mice gravitate to temperatures of between 30 and 32 degrees Celsius (the equivalent of about 86 and 90 degrees Fahrenheit). But based on federal regulations, U.S. research laboratories are routinely kept on the cold side -- between 20 to 24 degrees C. There can be advantages to these cold temperatures. For instance, mice have aggressive tendencies that are suppressed in cooler climes. Female mice also lactate better in cooler temperatures, though their pups don't do as well in the cold.

When kept in temperatures toward the low end of this scale -- between about 18 and 20 degrees C (64-68 degrees F) -- the mice begin to show changes in immune function and their growth may be retarded. "So we're housing them right at that threshold," Garner said. "That means the mice may be compromised physiologically, potentially affecting research results."

Simply raising the temperature in the lab isn't an option, not least because the mice would then become unmanageably aggressive, he said. Rather, Garner and his colleagues looked to other options in their study, which involved 36 male and 36 female mice of three common strains. The researchers created sets of two cages linked by a small tube so the mice could move between them. One cage in each set was maintained at a chilly 20 degrees C (68 degrees F) and was equipped with varying quantities of shredded paper, which the animals could use to construct nests for shelter and warmth. The other cage was kept at one of six temperatures: 20, 23, 26, 29, 32 or 35 degrees C (68, 73, 79, 84, 90 or 95 F), but without nesting material.

The mice then had the choice of staying put and tolerating the cold, choosing a balmier cage, eating more to add fat and elevate metabolic rate, or building a nest.

Each strain and sex had slightly different preferences, the researchers found. None was content to simply sit out the cold, either moving to a toastier location, if available, or building elaborate, dome-like nests to warm themselves. The more nest-building material they had, the more they were willing to settle for a cooler clime, as the nests served to temper the chill, the researchers found.

In fact, the nest-building drive was so strong that the mice often would spend hours collecting strands of paper, bit by bit, from the chilliest cage and then transporting it to a more comfortable spot in another cage to build a sturdy little home.

Garner said these mice decided they wanted to have it all, choosing a warm spot and building a nest as well. "Naughty little rascals" is how he described them. "They would go on holiday somewhere warm AND take their nest with them," he said. "Some people like to take a pillow on holiday and some don't. These mice were packing their own pillow."

The fact that some mice moved nesting material to the warmer cage means that the nests serve a function beyond warmth, argued Garner, perhaps providing physical comfort, or a form of protection that decreases the animals' anxiety and stress levels.

The nest-building mice tended to eat less, as they didn't need the extra calories to satisfy their higher metabolic demands, the researchers found. In general, the females preferred warmer temperatures than the males -- by about 5 degrees: they are smaller and have less fat to generate heat.

The researchers concluded that the mice could manage with 6 grams of nesting material but sometimes could use as much as 10 grams, suggesting the larger amount be supplied routinely in research labs.

Another benefit of the nests is that they facilitate researchers' work with the mice -- it's easier to pick them up as well as observe them. "The shape of the nest tells an experienced person whether the animals are too hot or too cold, whether they are sick or whether they are about to give birth," Garner said. "Once you learn how to 'speak mouse nest,' the nest is a wonderful tool that anyone can use to assess the general state of the mouse."

The study's first author is Brianna Gaskill, PhD, at the Charles River Laboratories. Garner and Gaskill collaborated with colleagues at the University of Calgary, Purdue University and the Environmental Protection Agency. The study was supported by a grant from the Universities Federation for Animal Welfare, the Charles River Laboratories (which donated the mice used in the study) and Fiber Core, which donated the nesting material.

According to the survey, Communication Skills ranks as the highest quality that employers want. This came out in 85% of the responses.

Tips and Commentary: When we talk about communication skills, most applicants immediately think that it?s about English Proficiency. This is mostly true as English is the preferred language in the field of business and the corporate world. Many companies look for people who can read, write and speak English very well, especially if they have international clients.

However, let?s dig deeper and find out what Communication Skills is really about. Communication Skills is not just about writing and speaking English well. It encompasses reading, listening, writing and speaking ability. It is about how well you can understand and interpret what you hear and read, as well as how good you are at getting your message across in such a way that other people can understand you.

To improve your Communication Skills, I suggest reading as much books, newspapers and articles as you can. Discuss these with others so that you can test yourself if you truly understood the message, while you practice getting your message across. If English proficiency is your weakness, use the English language more often. If it?s another language, do the same. Practice, practice, practice! The only way you can improve your communication skills is for you to, well, communicate.

2. ATTITUDE, VALUES & PERSONALITY

The second most common characteristics that our respondents look for is attitude, values or personality. This came out in 72% of the responses.

Tips and Commentary: When we talk about attitude, values and personality, we are talking about a person?s principles, moral standards, ethics and even disposition that affect his or her behaviour in the workplace. Evaluating a person on these is not an exact science. Although there are measurements that can be used, most recruitment officers rely on their intuition, gut feel and judgement to determine if the applicant is fit for the prevailing culture of their company.

You have to understand that values and cultures may greatly differ from one company to another. Some companies put more importance to teamwork, other companies prefer individual achievements while others are very competitive in nature. Some companies encourage employees to be proactive with lean set ups, while some companies have bureaucratic environments.

To improve in this area, it is best to go with positivity since practically all companies want positive people in their company. You would also want to determine your identity as a person and your principles in life. When you know who you are, it will be easier for you to see if you will fit in a particular company. I do not suggest changing yourself drastically and giving up your principles to fit in, but rather, it is more important to find a work environment and culture that suits your personality and values.

3. INTERPERSONAL SKILLS

The third most common trait that our respondents mentioned is Interpersonal Skills. This came out in 53% of the responses

Tips and Commentary: When we talk about interpersonal skills, we are basically referring to people skills---your ability to interact socially with others. Most people mix this up with communication skills because both are part of the realms of communication.

However, let?s draw the distinction between the two. Communication Skills is the general umbrella where interpersonal skills would fall under. The basic requirement of Communication Skills is your ability to read, listen, write and speak a language well. People Skills, on the other hand, consists of being basically good at communicating but it is more than that. It reflects how well you are able to relate and work with people, which is highly important in any organization. This is where non-verbal communication such as facial expression and emotions come in.

To improve your Interpersonal Skills, learn how to smile. This may sound very simple, but it nevertheless is very important in the workplace. When I interview job applicants, even before I ask questions, the first thing I look at is the applicant?s facial expression. I?ve interviewed so many applicants in my lifetime and you would be surprised that there are many who have very limited smiles. Interestingly, when I talk to other managers who make the second interviews, they would tell me that the reason why they rejected a person is that he or she isn?t too smiley or not very pleasant. Other things you can do to improve interpersonal skills is go back and learn the basics---etiquette, good manners, respect, politeness, courtesy, ?pakisama? and ?pagunawa sa kapwa?.

4. TECHNICAL SKILLS / JOB KNOWLEDGE

The fourth most common trait that our respondents mentioned is Technical Skills and Job Knowledge. This came out in 49% of the responses

Tips and Commentary: When we talk about technical skills or job knowledge, we are referring to the competencies required for the particular job that you are applying for. Each job position requires your ability to do tasks and functions. For instance, if you are applying as a Customer Service Representative, the employer may include customer focus, problem solving, product and process knowledge as part of the competencies required. If you are applying as an Office Secretary, the employer may include computer literacy, typing speed, filing and documentation as competencies required for the Job.

To improve your technical skills and job knowledge, you may want to go for additional training on your particular field of interest. Try to learn new skills that will help you be more competitive in the job market. We are not talking about just getting a certificate or a degree although that would help, but rather learning how to do things, understanding concepts and applying them on the job. However, if you do not have the time and resources to pursue formal training, the least you can do is study the job requirements and specifications and prepare well for the interview, preferably mentioning these to impress the recruitment officer. This will show that you did not randomly applying at just any company, but you are interested in the job you are applying for.

For example, let?s say you see a competitor running ads in an industry publication, so you decide you should run ads there too. The problem is you don?t know if those ads are working for the competitor or not.

Instead, use recommendations from trusted sources. Watch to see if promotions are repeated and appear on multiple channels (for example in print and online). Once you have done your homework, do a small test to see if the idea transfers to your business.

5) Test, Test, Test. Dan Kennedy gives this advice over and over and he?s right. Don?t wait until your marketing isn?t working. ?Continually test and be ready with the next promotion before you need it.

When it comes to picking marketing strategy that will win you more customers, clients or patients, remember not to leave your emotions behind and not get too attached to any one method or idea.

The 'Rich List' kept getting richer in 2011

They say that wealth begets wealth. That sure rang true for many hedge fund managers in 2011.

The ?Rich List? of Wall Street hedge fund managers, who took home a combined $14.4 billion last year, includes businessmen who culled their wealth in part through fees charged to pensions and to manage money for the wealthy.

The annual ranking by AR Magazine, an industry publication, also includes managers who made millions even though the returns on their funds?didn?t do much better than the overall S&P 500?s performance last year.

And in a year when the Occupy Wall Street movement protested inequitable wealth distribution, top hedge fund managers handling investments for the super-rich still found ways to make big money.

Take for example Ray Dalio, founder of Bridgewater Associates, who earned almost $4 billion mostly by betting on U.S. Treasuries, according to a TODAY report Friday by NBC senior investigative correspondent Lisa Myers.

?My customers are pension funds, teachers. I did well when others didn?t and I?m going to say that they are very grateful,? Dalio said in an interview with PBS.

Two years in a row, Dalio's firm earned more than Google, Yahoo, Amazon and Ebay combined, Myers found. And Dalio?s earnings were just considered ?pretty good? by industry standards.

?The industry?s fees and performance are so out of whack it?s unbelievable,? Bradley H. Alford, a former hedge fund investor who now oversees a mutual fund firm told the New York Times. "Fifteen years ago, you got double-digit performance for those returns, but last year, the S&P was positive and hedge funds were negative. There?s no alignment with fees.?

Number 2 on the list, shareholder activist Carl Icahn, raked in $2.5 billion.

Number 3, James Simons, who owns a stake in the Renaissance Fund, earned $2.1 billion. Citadel hedge fund head Ken Griffin got $700 million.

Griffin has a taste for the good life, Myers found.

His wedding reception was held at the palace of Versailles. He owns at least four homes in the U.S. including a $17 million home in an exclusive Hawaiian resort. He donated an eponymously named wing to the Art Institute in Chicago.

Then there?s the pauper of the top five: Steve Cohen.

Cohen, head of SAC Capital Advisers, earned a comparatively paltry?$585 million. He?s also a big spender.

He has?a 35,000 square foot mansion, owns a piece of the Mets and his world-class art collection is worth millions.

There were some losers in the group. Last year?s standout star, hedge fund manager John Paulson, founder of Paulson & Co., is believed to have personally lost about $3 billion ? losses partly attributed to his investment in a Chinese timber company later accused of fraud, according to the New York Times.

All?these billionaires either declined to comment or did not respond to NBC's calls. The managers argue that, unlike some corporate CEOs, they only do well if their investors do well.

Dalio has pledged to give away at least half of his wealth to charity.

Florida Quiet Title Actions: 10 Things You ... - About Florida Law

As the Florida real estate market stumbles along, we are predicting that land title issues are going to be causing more and more problems for Florida home owners, those wanting to sell Florida property, as well as those who want to buy a Florida home, condo, or tract of land.? Why?? The ForeclosureFraud mess has disrespected and ignored longstanding Florida laws designed to protect real estate titles over time.

The integrity of the system has been harmed, and it will take cleaning up lots of titles to get things back where they need to be.? Everyone involved in Florida real estate should proceed with care in dealing with real estate title transfers (sales, purchases, liens, insurance, etc.).

Here are ten things that everyone needs to know about Florida land titles and the process of quieting title to land and real estate here in the State of Florida:

1.? What is ?title?? When someone owns ?title? to land in Florida, it means that they have some form of legal ownership in that property.? In Florida, the Florida Legislature and the Florida Courts are very, very careful to protect the recording of land ownership and the process for how real estate is transferred from one owner to another.? This is the title process.? For example, here is a law that was passed long ago ? in 1828 ? that is still effective today:

689.01 How real estate conveyed.

No estate or interest of freehold, or for a term of more than 1 year, or any uncertain interest of, in or out of any messuages, lands, tenements or hereditaments shall be created, made, granted, transferred or released in any other manner than by instrument in writing, signed in the presence of two subscribing witnesses by the party creating, making, granting, conveying, transferring or releasing such estate, interest, or term of more than 1 year, or by the party?s lawfully authorized agent, unless by will and testament, or other testamentary appointment, duly made according to law; and no estate or interest, either of freehold, or of term of more than 1 year, or any uncertain interest of, in, to, or out of any messuages, lands, tenements or hereditaments, shall be assigned or surrendered unless it be by instrument signed in the presence of two subscribing witnesses by the party so assigning or surrendering, or by the party?s lawfully authorized agent, or by the act and operation of law. No seal shall be necessary to give validity to any instrument executed in conformity with this section. Corporations may execute any and all conveyances in accordance with the provisions of this section or ss. 692.01 and 692.02.

2.? Different Ways to Take Title. In Florida, you can own real estate in several different ways.? Under Florida law, you can ?take title? to real estate through any of the following ways? and which option you choose may depend upon several factors: estate planning may be a consideration, for example, and tax issues can be another.

Sole Ownership ? here you take title to the real estate in your name.

In Trust ? in Florida, land title can be held in the name of a trust, for the benefit of the trust?s beneficiaries, including a? Living Trust and a Florida Land Trust.

Shared Ownership ? here, you take title to the property with one or more other people as joint owners.? Two examples: land title held as ?tenants in common? or ?joint tenants with the right of survivorship.?

3.? Who Can Hold Legal Title to Florida Real Estate? Floridians, of course: residents of the state own most of Florida real estate.? Americans living in other states can also hold title to Florida real estate as well as citizens of other countries.? In fact, buying pretty Florida vacation spots is very popular right now with people all over the world (Canada, Great Britian, Brazil, etc.).

However, you don?t have to be a real, living person to hold legal title to land here in the State of Florida.? Corporations and partnerships can hold legal title to Florida real estate.? So can the local, state, and federal governments.

4.? What is the process of clearing title to Florida real estate? Making sure that you have clean and clear title to real estate is done by a ?quiet title? action, which is a lawsuit that is filed in a Florida court.? Getting a title insurance policy protects you against claims by others who are asserting a right to your property, but a title insurance policy does not clean up legal title.

5.? What is a? quiet title action? A quiet title action is a lawsuit filed in a Florida circuit court.? It is a civil action.? You are the party bringing the action, and you are asking the judge to issue a judgment.

6.? How long do quiet title actions take? How long the lawsuit will take, from start to finish, depends upon the circumstances of your individual case.? However, there is the advantage that in Florida, this lawsuits proceed in circuit court and not district court.? Florida district courts are bottle-necked and clogged with all the foreclosure lawsuits filed by the banks and mortgage servicers. Quiet title actions aren?t hampered by that backlog.

7.? What Does a Quiet Title Action Accomplish? A quiet title lawsuit results in a judgment, signed by a Florida judge, that can be filed in the real estate records to demonstrate that the judge has ruled and declared that all adverse legal interests to your property have been removed.? The chain of title is clear on the land, from you as the current owner, going back in time via the real estate records.? The title has been ?quieted.?

8.? What Is Left After a Quiet Title Action? A quiet title action will not erase legal claims to the real estate that are shown to be legitimate under the law.? For example, if there is a Federal Tax Lien on the property, that may survive the quiet title action.? Similarly, as discussed in our last post, a bank?s mortgage lien may survive a quiet title action.

9.? What is a Chain of Title? The chain of title on your land is the historical record of who has owned that tract, from the current day back in time as far as the records can go.? Real estate records are kept at the clerk?s office that track the land from today?s owner back over the years.? There are places in Florida where title investigations can go back into handwritten land records on parchment (these have usually been scanned, microfiched, etc.).

If you have a particular tract of land that you are interested in investigating, you may be able to start your quest right now, from your keyboard.? Go here and follow the instructions of the Florida Department of Environmental Protection (DEP), Division of State Lands, Board of Trustees Land Document System.

10.? What is a ?Cloud on Title?? When the chain of title for a particular piece of real estate isn?t straightforward and there is either a gap, or there is an adverse interest making a claim of ownership to all or part (e.g., mineral rights, air rights, water rights, joint tenancy, etc.) in the property, then the chain has a cloud on it.?? Any kind of cloud on a real estate title must be removed in order for you to own your land free and clear.

Larry?s Tip

Quiet title actions are not complicated in the procedure to be followed; these types of lawsuits have been honed over the years till they are familiar to most Florida real estate attorneys.? The problems that will be happening now and in the future are the complexities that the Wild West Mess of Foreclosure Fraud, robosigning, resale of mortgages in mortgage securities, etc. create.

In the past, a quiet title action might be brought because a title deed had a typo in the legal description of the property, and the wrong block number or condo unit was shown.? Easy as pie, a judgment was entered to fix the typo.? Problem solved.

Now, quiet title actions will be more adversarial because of the mess that has been created by so many institutions disrespecting real estate law.? The integrity of the Florida land title recording system is at stake, and there will be many a courtroom battle to win that war.

Benitez pitches Premier League clubs for new job

MANCHESTER, England (AP) -Lecturing football executives on the art of management, Rafa Benitez made a public pitch on Wednesday to be given the chance to work in the Premier League again.

Despite being out of work for 15 months, however, the former Liverpool manager made it clear he will only accept an offer from an English club that has enough money available to potentially become title winners.

Benitez had been a contender to replace Andre Villas-Boas at Chelsea last month, but no formal offer was made. Roberto Di Matteo was promoted to manage the team until the end of the season.

"At this moment my priority is to stay in the Premier League, and I know that I can win. With my people and with my team, we can win trophies," Benitez said at the SoccerEx convention. "When you have been at the top winning trophies, you have a lot of people working with you and you have a methodology - you think you can be successful again."

Benitez has been out of work since being fired by Inter Milan in December 2010, only six months after the end of a six-year reign at Liverpool in which he won the Champions League and FA Cup.

"I am not desperate because I have had a lot of offers," Benitez said. "But I am looking for a new job."

Liverpool has struggled since Benitez left after the 2009-10 season, with current manager Kenny Dalglish under pressure despite winning the League Cup last month as his team is seventh in the Premier League.

Benitez still lives in the Liverpool area, adding to suggestions he could return to Anfield one day.

"There is no rush and if Kenny retires in 10 years' time and I am available, I will be really pleased," Benitez said. "Not now because they have a manager with the support of the owners."

For now, Benitez is promoting his coaching aid application for smart phones and computers. His SoccerEx presentation veered from being a sales pitch to reminding executives in the audience about his managerial credentials.

Responding to a question from the audience, he downplayed the significance of engaging in a war of words with rival managers in public.

"I don't think it's important or makes a big difference," Benitez said. "You can win all the mind games but if you don't have a good team, you can't play mind games ... when you have a lot of foreign players, they don't usually read the papers. So the mind games for them are nothing.

"After all this work behind the scenes. you don't need to be worried about the mind games. You need to be worried about your job and the game."

Stanton Peele: The Constitution and Health Insurance

Are there ways in which the Constitution isn't the inerrant guidelight we were taught to think of it as in secondary school? That is, what about airplane security?

But I am thinking specifically about health care. As Glenn Beck so notably argued, "The Constitution doesn't guarantee health care as a fundamental right."

Right. But it has become awfully important to us as Americans and as human beings. And, damn it, they get it as a basic right in other countries! Are they better off than we are because they don't have a Constitution?

Having introduced that question, I'll now leave it for Constitutional scholars to ponder.

But what I'm really thinking about is how we argue the entire heath care debate in terms of personal choices (which may be violated by a mandate to purchase insurance), the rights of businesses and free enterprise (as in, let insurance companies compete to make us the best offers from which to choose our health plans), and a kind of you're-on-your-own, personal freedom philosophy that our founding fathers thought in terms of, but which now seems largely to benefit some very rich and powerful people who control much of all of our lives.

Which leaves us -- where? As a nation, wanting more secure access to our wonderful doctors for ourselves and our children, anxious about how we can afford insurance (or, as seniors, which I am, about how much Medicare covers). Quite understandably, we want more coverage, and to pay no more (please God, maybe even less?) for it.

But personal health care costs continue to rise sharply for everyone, more so than they do in those countries without Constitutions.

As one example of an alternative universe, consider New York Times columnist Thomas Friedman's recent description of health care in Australia and New Zealand: "Conservatives in Australia and New Zealand have also long accepted single-payer national health care systems." Friedman explains:

There are many reasons for the narrowness of the political spectrum here, Johansson added. Neither New Zealand nor Australia are strong churchgoing countries, so social issues don't resonate as much. Both being isolated, sparsely populated, pioneering communities -- New Zealand has only 4.5 million people -- they have strong egalitarian traditions and believe the state has a role to play in making sure everyone gets a fair shake.


"We also have compulsory voting," said Turnbull [Malcolm Turnbull, former leader of Australia's main conservative party]. You get fined if you don't vote. "In a voluntary voting system like yours, there is always the temptation to run hard on hot-button issues that will fire up the base and get them out to vote. In a compulsory voting system, your base has to vote -- as does everyone else -- and so the goal is to target the middle ground."

Wait a second -- doesn't that last bit violate the Constitution?

We're in a sad situation, our American lot, many of us a bad illness or an accident or a denial by an insurance company away from losing our precarious balance in life.

And is this all written in the Constitution?
