Ovary stem cells can produce new eggs, researchers say
If Sunday's report is confirmed, harnessing those stem cells might one day lead to better treatments for women left infertile because of disease -- or simply because they're getting older.
"Our current views of ovarian aging are incomplete. There's much more to the story than simply the trickling away of a fixed pool of eggs," said lead researcher Jonathan Tilly of Harvard's Massachusetts General Hospital, who has long hunted these cells in a series of controversial studies.
Tilly's previous work drew fierce skepticism, and independent experts urged caution about the latest findings.
A key next step is to see whether other laboratories can verify the work. If so, then it would take years of additional research to learn how to use the cells, said Teresa Woodruff, fertility preservation chief at Northwestern University's Feinberg School of Medicine.
Still, even a leading critic said such research may help dispel some of the enduring mystery surrounding how human eggs are born and mature.
"This is going to spark renewed interest, and more than anything else it's giving us some new directions to work in," said David Albertini, director of the University of Kansas' Center for Reproductive Sciences. While he has plenty of questions about the latest work, "I'm less skeptical," he said.
Scientists have long taught that all female mammals are born with a finite supply of egg cells, called ooctyes, that runs out in middle age. Tilly, Mass General's reproductive biology director, first challenged that notion in 2004, reporting that the ovaries of adult mice harbor some egg-producing stem cells. Recently, Tilly noted, a lab in China and another in the U.S. also have reported finding those rare cells in mice.
But do they exist in women? Enter the new work, reported Sunday in the journal Nature Medicine.
First Tilly had to find healthy human ovaries to study. He collaborated with scientists at Japan's Saitama Medical University, who were freezing ovaries donated for research by healthy 20-somethings who underwent a sex-change operation.
Tilly also had to address a criticism: How to tell if he was finding true stem cells or just very immature eggs. His team latched onto a protein believed to sit on the surface of only those purported stem cells and fished them out. To track what happened next, the researchers inserted a gene that makes some jellyfish glow green into those cells. If the cells made eggs, those would glow, too.
"Bang, it worked -- cells popped right out" of the human tissue, Tilly said.
Researchers watched through a microscope as new eggs grew in a lab dish. Then came the pivotal experiment: They injected the stem cells into pieces of human ovary. They transplanted the human tissue under the skin of mice, to provide it a nourishing blood supply. Within two weeks, they reported telltale green-tinged egg cells forming.
That's still a long way from showing they'll mature into usable, quality eggs, Albertini said.
And more work is needed to tell exactly what these cells are, cautioned reproductive biologist Kyle Orwig of the University of Pittsburgh Medical Center, who has watched Tilly's work with great interest.
But if they're really competent stem cells, Orwig asked, then why would women undergo menopause? Indeed, something so rare wouldn't contribute much to a woman's natural reproductive capacity, added Northwestern's Woodruff.
Tilly argues that using stem cells to grow eggs in lab dishes might one day help preserve cancer patients' fertility. Today, Woodruff's lab and others freeze pieces of girls' ovaries before they undergo fertility-destroying chemotherapy or radiation. They're studying how to coax the immature eggs inside to mature so they could be used for in vitro fertilization years later when the girls are grown. If that eventually works, Tilly says stem cells might offer a better egg supply.
Further down the road, he wonders if it also might be possible to recharge an aging woman's ovaries.
The new research was funded largely by the National Institutes of Health. Tilly co-founded a company, OvaScience Inc., to try to develop the findings into fertility treatments.

As more and more people are finding themselves in long distance relationships, the belief that it is bound to fail has been proven to be unsubstantiated.
Many are in fact bound to flourish. I know from personal experience. I jumped in heart first to a long distance relationship nearly three years ago, and that has blossomed into what will soon become a marriage.
Over the past three years we have had times spent together and apart in three different countries. We have had times of intense connectedness and times of utter despair, but most importantly together we have learned how to use the ebb and flow of distance to our advantage.
Like other relationships, there are an infinite number of things that play into the success of a long distance relationship. I can offer ten tips that I wish someone would have shared with me. These won?t guarantee success, but I can assure you that they will make your long distance love life more about the two of you and less about the distance in between.
Although this distance may seem daunting at first glance, my hope for you is that you can soon see the challenge as an opportunity to grow individually and with your partner.
- Honesty must be your first policy. By that I mean you need to be honest with yourself and with your partner. Long distance relationships offer no room for guessing games of hard to get or otherwise. Be honest about your feelings regardless of how irrational they may seem. Tell each other where you stand in the relationship. That means both people know if the relationship is open, closed or on hiatus. This is even important for any lengthy separations.
- Set appointments. Whether it be for skype sessions, phone calls or however you communicate with each other, this is important. It gives each partner something to look forward to and prevents any arguments over why one person didn?t call or answer. This also means that you need to be on the same page about how often you want to talk. What each person expects may be different, so find a compromise.
- Make an effort to actually see each other. Even if it is just some sort of video chat once a week. We are very visual creatures. Seeing each other can really kindle the fire between the two of you.
- Continue to share your common interests. This can be anything. Is there a website that you both visit regularly? A show that you both watch every week? A sport that you both enjoy? A movie you both want to see? Maybe it?s a book, a class, an artist. The point is that you keep doing the things that you like to do when you?re together, even when you are apart. It gives you something to talk about when the conversation wanes and more importantly, you can feel connected to your partner despite the lack of physical presence.
- Do something unexpected. This could be something as fun as a surprise visit (provided your relationship has reached ?surprise visits are okay? status) or as simple as handcrafted snail mail, which is my personal favorite. Text messages are also a sweet way to be reminded that there is someone special in your life. Be creative and find what works for you and your partner.
- Discuss your expectations. Especially your expectations regarding the future of the relationship. Expectations have the potential to ruin any relationship if they are not considered thoughtfully and kept in check at times.
- Be realistic about when you will see each other next. This is really simple, but it?s sometimes forgotten when you?re desperate to see each other again.
- Enjoy your time. Long distance relationships are not for the faint of heart. You need to really be able to examine yourself as an individual. Stay involved in your own life, after all this is most likely one of the reasons your partner was attracted to you in the first place.
- Accept bumps in the road. They will come. If you are not resilient and truly committed to the health of the relationship, you will give up easily. Remember that you will feel lonely and not connected at times. Rest in the fact that feelings are transient in their nature.
- Talk to each other. Of course, you are going to talk to each other on the phone, through email or otherwise, but I?m referring to talking when nobody is around. Yes, this sounds a little crazy but it does provide some comfort just to express yourself out loud. Plus, it?s better than just talking to yourself.
As for the rest, experience is the best teacher. There are many successful long distance relationships out there, and I?m happy to be part of that group.

[A]List Game Marketing Summit: Pachter's Bold Keynote - [a]listdaily
Michael Pachter, Managing Director of Equity Research at Wedbush Securities, gave the keynote address at the [a]list game marketing summit in San Francisco on Thursday, and he had a number of provocative observations for the assembled crowd of game marketers.
Pachter lead off with his prediction for the industry standby of packaged console products.?I think console packaged products are going to be around for a long, long, long time,? said Pachter ?When they do go away, they're going to be replaced by console download products. People are going to have that $60 ? soon to be $70 ? game play experience as long as there's somebody who has a vested interest in making sure that happens.?
Pachter took exception with an observation by David Cole that was cited by Steve Fowler of the Ayzenberg Group, who introduced Pachter and the summit. ?I think David Cole said something that's wrong. He said there could be a social bubble, because all the games are copycat, there's no originality... you could absolutely say that about first-person shooters. You could. They're all the same game. Except they're not. I would say that about Hidden Chronicles and Gardens of Time. I love Zynga games, and I get why they're doing them,? Pachter said. He followed with double-edged compliment: ?Zynga actually does a great job with their games. Even when they rip somebody off, they do a better job than the person they ripped off.? That's one quote that Zynga probably won't be using in their marketing materials.
Pachter went on to compare the evolution of the gaming industry to the evolution of the casino gaming industry. Pachter noted that to be a successful player, games like poker require a deep knowledge of the game and a lot of play time to develop the necessary skills. (He also said that he paid his through law school as a poker player, earning $250,000 per year for three years of law school.) Poker is played in long sessions; you don't just sit down and play for a few minutes, Pachter noted. Casinos all have a poker area now, but they also have table games like craps and roulette, which are kind of complicated but not as highly skilled as poker. You spend time playing them, but you don't have to spend as much time as when you play poker. The biggest area in casinos is devoted to slot machines, which anyone can sit down and play for a minute or two or for hours.
How is all of this like the gaming industry? Pachter explains: ?Texas Hold'em is like playing an MMO. Exactly like one. Nobody does it for five minutes, and when you play it you are immersed in it. The same kind of people do both, obsessive-compulsive people.? In Pachter's analogy, the table games are console games. ?You cannot just start playing Call of Duty. You tend not to play it for a few minutes,? said Pachter. In both cases you have to learn something about the games and develop some skills, and you tend to play them for at least an hour or two.
Then you have slots, which Pachter says ?are just like casual, social , and mobile games. Everyone can figure them out, and they're really mindless.? When Las Vegas was just table games, that was like the game industry with just console games. ?Console games were really like Vegas thirty years ago. The 90s was when the MMO started to appear, and that's when the poker rooms started to appear in casinos,? said Pachter. Now we see that casinos have devoted a huge amount of space to slots, and they generate a lot of profit for the casinos because they are so easy to play for anyone, and you can play them for a minute or for hours. The casinos still have the table games, and they still have the poker rooms, and they'll continue to draw a good audience even though the slots may get the majority of the floor space.
Pachter now sees social games as a good business that will continue to grow, especially when the social game companies are able to refine their business model. ?Free-to-play is free, and that's a mistake,? said Pachter. ?Free to play should not be free. In Western cultures, we get nothing for free. It's ad-supported if you don't pay for it.? He pointed out that we have no problem watching an ad in order to listen to music on the radio, or to watch television, or see movies on TV. The scoial game market needs to take that to heart, according to Pachter. ?If publishers and developers can figure out a way to monetize the 98 percent of the people who don't pay, this is a great industry to work in.?
Pachter had quite a lot to say about the console market, too. ?Smartphones have absolutely destroyed the casual market for consoles. They also killed the casual Wii game,? Pachter noted. ?The market for hardcore games is bigger today than it's been. Nintendo inflated the numbers with a bunch of great casual experiences, which have been replaced by social games, and smartphone games, and tablet games. I can tell you that the women that were playing the Wii Fit are now playing FarmVille, CityVille, CastleVille. Wii was a bubble. The Wii bubble has burst.?
We took advantage of the question period to ask about the next generation of consoles. New consoles are in the wings, the Wii U's been announced, but pricing hasn't been announced. There are rumors that Microsoft's next console might be as much as $600. Could the next generation of consoles really be successful at that kind of price point?
Pachter didn't hesitate to share his thoughts on the next generation of consoles. ?Since Microsoft hasn't settled on specs, I think that's probably premature,? said Pachter, referring to the idea of a $600 price point for the Xbox 720. He moved on to his thoughts on the Wii U. ?I think Nintendo's in disarray. I think the idea that we don't know the price point yet, but we do know what the console is, is just sad. I think they've completely blown that. It's gonna launch at $249; because it has to. They're dead if they launch at $259, I think they're toast then. I think they're toast anyway.? Why? ?I think the Xbox 360 with Kinect will be priced below that by the time they launch.?
Pachter went on to talk about the next console generation in general. ?I'm a little worried about the next generation because I actually think we've gotten to the point where people don't care any more. Look at television displays; we're at 1080p (2K) already and they're already talking about 4K and then 8K. Nobody's buying a 4K TV or an 8K TV in the next ten years. They're $15,000! All the consoles now are running in 1080p, so what are we gonna get? 60 frames per second standard, and 120 frames per second occasionally? Who cares? Have you played NBA 2K? It looks like a frickin' movie. The facial animation isn't great, but the game looks like a movie. It doesn't get any better than that. I'm actually worried about them in general.?
Pachter finally circled around to the pricing question. ?So the answer to your question is, yeah, price I think is everything, because I don't think features matter that much. I think what you're going to see in this next generations of consoles is this: 4 GB hard drive, except for Nintendo which will be no hard drive, and maybe some type of built-in cell access or something, some way to connect with the television without wires, it'll be like little features... but as far as gameplay, you're talking about faster framerates, but I'm not even sure what manufacturers want. We're in a really interesting time. I think the manufacturers better figure it out and better let consumers know, because we don't want to make them to wait for something that's not coming. If Microsoft launches at $600 it's over.?
Nintendo's pricing of the Wii U will be a very important part of the puzzle; not just the price, but when they choose to announce it, and then how Sony and Microsoft respond. The holiday market in 2012 will be very, very interesting to watch.

New moth invades Italy vineyards
A moth with a taste for Chardonnay leaves, which has infested vineyards across northern Italy, is a new species of leafminer, scientists say.
The pest was first discovered by Italian scientists in 2006, but they were unable to identify it.
Now, by examining a snippet of the moth's genetic code, researchers have confirmed that it is a previously unnamed species.
The team published their findings in the journal ZooKeys.
The Italian team enlisted the help of insect expert Erik van Nieukerken from the Netherlands Centre for Biodiversity in Leiden.
"We first turned to the [scientific] literature to find out what was already known, which was appallingly little for this group [of moths]," Dr van Nieukerken told BBC Nature.
He and his colleagues used a method known as DNA barcoding to examine a section of the insect's genetic code.

Only the genetic studies revealed it to be a different species with a taste for grapevines. Its native range is across eastern North America, where it feeds on several species of wild grapes.
So far, the species has been found in vineyards in Italy's Trento and Veneto regions, spreading and increasing in population since it was first recorded.
Having observed the moths in the field, the scientists say that the insect seems to have a preference for the leaves of Chardonnay, Cabernet Sauvignon and Muscat grapes, but they added that the economic impact of this particular pest was not yet clear.
The researchers do not know exactly how the moth arrived in Italy, but Dr van Nieukerken said that it was very easy for the cocoons containing the larvae to be accidentally transported with plant material.
"They're very small and exactly the same colour as the leaves," he told BBC Nature. "So if you were carrying plants, you would probably not notice them.
The scientist said that another species from this same group had been discovered in commercial walnut crops and that more needed to be known about the insects.
"This group is very poorly studied," he told BBC Nature.
"If you know exactly what it is and where it belongs, if you know its evolutionary history.... you can understand better how to control it."

Since it was the first time I had delivered my ?style? of lecture to a straight-up Evolution class, I wanted to be sure that my lecture was relevant.? I decided to focus on clarifying oft-confused concepts in Evolution. After a brief discussion with him, I decided to tie into the concepts he had recently lectured: Natural Selection, Adaptation, and Population Genetics.
I kicked things off with a discussion of the phrase Survival of the Fittest and lead off with lecture notes in my Hip Hop Evolution Files: Quick & Dirty explanation of Natural Selection by Daft Punk post.? Taking Karen James? critiques into consideration, I went into a full-on explanation of Lamarckian Evolution and the fallacy of conflating individual effort as contributing to adaptation.
Lamarckian selection focuses on the individual and the individual can ?shape? the traits in question. Unlike Larmarkian selection, natural selection focuses on the population and acts on variation of the traits.? Which I emphasized to students are phenotypes (P). Despite the heavy attention many scientists pay to genetic inputs of traits, Genes (G) are only half of the story ? if that much. I?m on team Environmental Effects (E).
But no doubt, individuals and their efforts still matter. After all, if an individual dies before it leave offspring ? for whatever reason ? then it affects what is in the collective gene/phenotype pool for natural selection to work with. You understand? Which brings us to another evolutionary biology tongue-twister: Heritability and Heredity and distinguishing Heritable Traits and Inherited Traits.

Heredity is with the likelihood or probability of traits running in families.? However, it is important to keep in mind that a trait can be heritable due to genetics (G) or environment (E) ? that whole nature vs. nurture argument. Environmental or nurturing effects include habits, behaviors, as well as physical/emotional/psychological experiences.? Relatives often demonstrate similar habits and behavior and we share experiences with them as well.
Genetically we share half of our genes with each of our parents and a quarter of the same genes as each of our grandparents.? These genes we get directly from our parents/grandparents, we inherit from them, as are the traits we have that are determined by these genes.
However, our shared habits, behaviors and experiences are not genetic, but are still heritable traits ? there are is a great likelihood we share these traits with our relatives. ?Heritability is the likelihood of getting something?in other words, you might get it. It?s not automatic ? like genetic inheritance, but the chances look good.? The important thing to keep in mind is that inherited traits are directly passed down from parents to children, whereas heritable traits are not necessarily genetic.
To wrap things up, I had students take a quick little quiz to see if they could distinguish between inherited traits and heritable traits.? Let?s see how you do. Play the first verse.

Depends. If he means she may be mentally ill it could be inherited. A few mental disorders are due to genetic mishaps, but I?m guessing he means her general way of behaving in public.? She has good or bad home training, therefore it is heritable.
Where does she get her income? She get it from her mamma.
Could be both. In human cultures, offspring often INHERIT wealth from parents, passed down directly from one generation to another. Of course this isn?t genetic, but the rule of ?being passed down directly from parent to child? means that it is inherited. However, income is also heritable, in the sense that often children follow the same occupation choices as their parents.

Strongest Long Distance Relationships ? reginecelinaaa

It?s Wishful Wednesday today! It?s also Valentines Week and since I?ve been posting about Love this week, I believe that it?s time to finally publish my special post about it. Mkay, so around September 2011, I made this blog post but I never published it yet. I just kept waiting for the right time. I initially made this because a friend of mine started being in an LDR and I wanted to let her know that there?s nothing she should worry about. I present this as a Valentines Week feature. Just because Valentines Day finished does not stop people in love from celebrating Love everyday!
Long Distance Relationships are honestly difficult but not entirely impossible.?Each relationship is different from each other but there are also some similarities that each may share. Each partner also possess different point of views on some things which is why I?ve asked four couples three questions:
- What are your thoughts and experiences on LDR?s?
- What is/was the hardest part of your LDR?
- What advice can you give to people who are experiencing or will experience LDR?s?
These are not your ordinary couples. These four couples are my friends that have experienced and/or still experiencing Long Distance Relationships. I actually have a lot of friends that are in an LDR but I chose these four couples because I perceive them to be one of the strongest out there. I look up to their?relationships?as an inspiration and a symbol of hope and true love. They?ve been through a lot and are as strong as you can imagine. These four couples are truly amazing. And here are their answers:

- I wasn?t able to get Kuya Mike?s side as he was busy.
Ate Sheila:
- At first I was hesitant to get into LDR because I?m well aware of its numerous disadvantages. Not to mention the ?stigma? in our society associated with LDR. Oftentimes i would come across with people whose opinions towards LDRs are mostly negative. These INFIDELS (that?s what I call them hehe!) would usually give unwanted remarks such as ?LDR??Is that even possible?? ?Who are you kidding?! For sure ur bf is cheating on u right this very moment!? ?c?me on be WITH me &I?ll get rid of Your loneliness. Since your bf is miles away from u? etc etc to say that LDR is not widely accepted, especially in the case of an unmarried couple is such an understatement that is full of ambiguity.?My initial reluctance was somehow justified by our countless and petty quarrels. Not just once have i asked for my ?freedom? such misunderstandings were often led by our inability to personally explain our sides. I won?t deny the fact that it was Michael who kept this relationship intact. To make LDR work, one or preferably both parties should be willing to endure any kind of pain imaginable. Sacrifice and a great deal of TRUST and UNDERSTANDING are highly necessary.
- Which part?? Well EVERY PART OF IT. Couples in LDR rely only with different means of communication. In our case chats, texting (he bought me a Smart roaming) overseas calls (regardless of the country?be it in England or even Bangladesh. I would come up with something to contact Michael) etc etc and ironically when you guys are finally together?that is even much harder?.why? Coz it?s like being offered all the sumptuous meals you could ever think of. Only because it?ll be your last as you?ll be executed hours later! So you see every minute that you guys are together is extremely precious that you would wish that it won?t end.
- Well now that we?ve literally come along way?. I can proudly say that LDR is INDEED POSSIBLE! It a matter of how steadfast and true is your love for each other. For me ?I love you? means ?I TRUST you?. Who ever said that love or the idea of being in love is ALL about joy and happiness is a lunatic and absurdly delusional! In order to fully understand and experience TRUE LOVE? one should be ready to face any kind of hurdles that he/she might come across. The willingness to accept that TRUE LOVE CAN BE SOMETIMES BE FULL OF SACRIFICES, ANGUISH & IS NOT ALWAYS IN YOUR FAVOR?is the crucial mind set of somebody who is ready to face anything in the name of love. And trust me, all these will bear fruits no materialistic luxury could ever surpass!

- A long distance relationship can either be the Achilles heel in your relationship or an opportunity for you and your partner to learn more about yourselves and each other. Its something that can either make you or break you. Based on my experience (so far) its been a way for me to appreciate my partner even more and a way for me to learn about my own limitations.
- Well, im sure Honey would tell you that the hardest part was during the beginning when the separation and the pain was still fresh. I knew that would happen. So i made sure i?d be strong enough for the both of us. You see, i cant really give you a straight answer to this question. Because a difficult time for one of you will eventually affect both of you. The beginning was hard for both of us? lets put it that way. There were (and still are) times when i really miss her and would like someone to talk to, not just anyone but someone that really gets me the way she does. Honey wasnt just my girlfriend shes also my best friend. So another difficult time for US is when there are things i want to tell her but cant. But most of the time, her damn internet connection always cuts out? needless to say, that can be really annoying!
- The best advice i can give you is to come into your LDR strong. What i mean by that is to have a solid relationship before even starting your LDR. In general any relationship that is founded on lies and insecurities will not last very long. So make sure you clear all this up before waking down this long and lonely road. Another important tip is DONT plan anything. The frustration will kill both of you if it doesnt work out, and believe me, in an LDR very few things work out according to plan. So just chill do what you need to do and go with the flow, learn to adapt to situations instead of reacting to them. And finally, FB, twitter, Google+ and Skype?. the internet is a world of its own and a great way to communicate with your partner. It might seem dorky at first but believe me, you?ll really appreciate it down the road.
Ate Honey:
- A lot of people don?t believe in it, but a lot of people have survived it as well. I think that as long as both parties are consistent in making an effort to make it work, then you should be fine. Keeping an open line of communication is one of the most important factors, as well as TRUSTING and HAVING FAITH in each other. Just believe that you?ll make it through and pray, pray, pray?.
- The hardest part was? the beginning. I would cry my eyes out every night. I cry when I hear a song that reminds me of us, I cry when I pass by a place we used to go to, I cry when I see his stuff lying around my room, I cry whenever I see our pictures together.. I cried all the time!! But I tried my best to stay strong for him because I know it hurts him too when I cry.. Kailangan tulungan kayo eh.. Because wala sya paghuhugutan ng lakas ng loob kapag pati ikaw sobrang depressed na. You?re supposed to be each other?s pillar of strength.?
?The hardest part nowadays? Its when our internet connection is being retarded. HAHA! Kidding aside.. Its when we have those fights that start out small and then biglang lalaki. Its frustrating because you?re not physically there to make lambing.?
- Besides the ones that I mentioned on the first question? What I can say is, before you get into a long distance relationship, be 101% sure na kaya mo panindigan yun papasukan mo. It?s not going to be easy so be sure that you can promise yourself and your partner that you won?t ever do anything to make him/her worry about you. Never put yourself in vulnerable situations like getting drunk with friends of the opposite sex, or doing something out of impulse when you get into an LQ. ALWAYS think of ways to make the other person feel that they?re always on your mind no matter how far you are from each other. Bawal ang hindi ma-effort sa LDR..?

??Misunderstanding will always be the cause of fight/argument, hindi maiiwasan yun. Especially if your schedule are conflict, pag uwi mo tulog sya, pag gising mo umalis na siya. Things like that have to be considered. What else, trust is very important anddd as much as possible, you have to update how your day went when you have the time to go online.?
Evan:
- ?LDR is a TEST. It tests how COMMITTED you are to the person. It?s a test of trust and most imporantly PATIENCE. If your partner have these abilities, then guaranteed, he loves you. From my experience, the longest LDR I?ve had with Hosni was around 1 year and 6 months of not seeing each other. If you want it in total, it?s approximately around more than 2 years. It was such a struggle during those times?the time difference, the no cuddling, the internet connection problems! But we struggled through all of those?TOGETHER?.online?:P
- The hardest part was when we broke up for a long time (6 months), when I?ve had enough of our nonsense fights. You know that after a couple fights, the LAMBINGAN goes next. We didn?t have that because we couldn?t do that together online! The only thing we did was talk on the mic with matching hugging and kissing pa in front of the webcam and it looks stupid if a 3rd person is watching you. Although it?s a good thing that we never longed for anyone else
- ?COMMITMENT + PATIENCE = You?ll get through it.

There are lots of highly anticipated events in February such as watching the Super Bowl, or getting a three-day weekend for President?s Day, or even finding out if the groundhog sees his shadow. But the most highly anticipated even would have to be the return of The Walking Dead
. I?ve been looking forward to its return since November. It might be my favorite show currently on television.
Oh, there is one other big event that happens in February. Valentine?s Day. My wife and I already had our Valentine?s celebration by staying in a hotel for a night and enjoying the hot tub and other amenities. We got a very good deal since the manager of the hotel is a member of the same business networking group as us. I think it was a much better way to celebrate Valentine?s Day than going out to eat on the actual day when everybody else will be out.
I didn?t make progress on my goals this week. My stepdaughter had surgery and was in the hospital a couple days and then we had our already planned pre-Valentine getaway. That didn?t leave me much time to get things done and I decided to just not worry about my goals this week. That can be a slippery slope but there are sometimes when life takes precedence over your goals.

New understanding of DNA repair could eventually lead to cancer therapy
A research group in the Faculty of Medicine & Dentistry at the University of Alberta is hoping its latest discovery could one day be used to develop new therapies that target certain types of cancers.
The discovery by Mark Glover, his graduate student Zahra Havali-Shahriari and post-doctoral fellow Nicolas Coquelle has shed light on what happens in cells when DNA is damaged. They solved the structure of a DNA repair enzyme called polynucleotide kinase/phosphatase, or PNKP. This allows them to see what is happening when this enzyme is repairing DNA.
Their findings have been published in the Proceedings of the National Academy of Sciences, a high-impact scientific journal.
In normal cells damaged DNA can lead to the breakdown of chromosomes and, ultimately, cancers. On the other hand, damaging DNA in cancer cells is a useful way to kill them. A long-term goal of this research is to find ways to specifically block PNKP from doing its repair work in cancer cells as a possible new cancer therapy.
"We can finally visualize it bound to the damaged ends of DNA," said Glover, a professor in the Department of Biochemistry. "We've trapped the enzyme bound to the damaged DNA before it actually repairs the damage. One of the surprising things that comes out of this study is that we also see that the enzyme has to unwind the DNA double helix."
Work over the last 10 years, pioneered in large part in the Faculty of Medicine & Dentistry, revealed that the enzyme PNKP plays a critical role in the repair of broken DNA ends produced by radiation and other agents. Until now, though, no one knew how it finds and repairs the damage.
"It breaks base pairs [of DNA] apart, peels off the broken end and then PNKP inserts that broken end into the enzyme," explains Glover. "It then performs a chemical reaction on the damaged DNA end, reversing the damage and releasing it so that the broken DNA strand can be welded together with the rest of the double helix. We now understand more about how this thing works; an enzyme that is protecting us from getting cancers."
However, the same enzyme also protects cancer cells. "We find a lot of tumours become resistant to these therapies [radiation and chemotherapy]," said Glover. "The holy grail of cancer therapy is to find drugs that we could give to people that would sensitize their tumours to these therapies.
"One way you could sensitize tumours is to target what they're using to repair damaged DNA. One of the ideas is that we could specifically inhibit this PNKP enzyme."
Sensitizing the tumours to therapies could also lower side effects, adds Glover.
The lab is already starting to test some compounds that could act as inhibitors for PNKP in tumours and they've seen some positive early results.
Because radiation is proven effective in some but not all cancers, new treatment avenues are necessary. Glover is playing a vital role in moving potential new treatment forward.
"It requires a lot of basic research to find out what's going on in all these different cancers," he said.

Lawsuit seeks to block Google's privacy changes
SAN FRANCISCO (AP) ? A consumer watchdog group is suing the Federal Trade Commission in an attempt to prevent Google from making sweeping changes to its privacy policies next month.
The planned revisions would enable Google to bundle the personal information gathered by its Internet search engine and other services so the company can gain a better understanding of its users and potentially sell more advertising.
But the Electronic Privacy Information Center contends Google's policy switch will violate restrictions imposed in an agreement reached with the FTC last year. A lawsuit filed Wednesday by the group is trying to force FTC to enforce those rules. The complaint also is seeking a court order to block Google's policy changes from taking effect March 1.
European regulators already have asked Google to delay the policy changes.
